Excited to share this recent preprint from the @MickLab. They cleverly join APEX-proximity biotinylation, which usually requires exogenous addition of H2O2, with D-aminacid-induced local H2O2 production by expression of the yeast DAO. It's genious!

Updated version of this manuscript is up now! We have new @zeiss_micro lattice-SIM data from our brilliant postdoc @JulienCicero showing how mito-associated actin bridges fusing mitochondria. We also have new data showing that mito-actin precedes MFN2 location to fusion sites 🤩
Alright I'm home sick with COVID and just submitted my grant, so I finally have bandwidth to do this now. So either buckle up or mute this thread (or both?!) cuz we're going on a journey combining brand new tools and new fundamental biological discoveries made with those tools.

Molecularly, the Pink1 mRNA tethering complex is phosphorylated by #AMPK to keep the amount of PINK1-mediated mitophagy low. Only upon untethering, e.g. upon inactivation of AMPK by insulin signaling, can mitophagy process and the neuron take care of damaged mitochondria.

This goes awry in neurons that have lost their ability to respond to insulin, e.g. upon treatment with the #Alzheimers disease (AD) risk factor ApoE4. This connects two phenomena commonly seen in AD, damaged mitochondria and #insulin resistance.

Tabitha identified a molecular switch that allows neurons to turn #mitophagy on and off, by tethering the Pink1 mRNA to mitochondria. This protects #mitochondria in neurons from being used as a simple nutrient source, because we all know they are much more important than that!
