Building T cell engagers that target multiple cancer signals | Founder & CEO Deck Bio | Stanford Immunology PhD

Cambridge, MA
Cancer isn’t driven by a single target. Our therapies aren’t either. We’re at #AACR26 this morning presenting DBXO-1, a multi-pMHC targeted T cell engager from Deck Bio. 📍 Poster #1632 (Board 24), Section 10 #TCellEngagers #ImmunoOncology #TumorHeterogeneity

$MRNA with the single most hilarious possible outcome in this study. EFS HR 0.58. Down the the hundredth, EXACTLY what *NEO*-adjuvant treatment with pembro yielded against adjuvant pembro (the nerfed comparator $MRNA studied against) in SWOG S1801. What does this mean in English? $MRNA’s drug is showing the exact same benefit versus adjuvant pembrolizumab as NEOadjuvant pembrolizumab did. Even more simply? If you just give patients a few doses of the pembrolizumab BEFORE you cut their cancer out, you do what $MRNA does for you, except with just the free tumor that’s already inside your body. No need to pay for this $400K drug. And this makes rational sense, right? The whole point of $MRNA’s vaccine is to produce stuff that looks like the tumor. You know what looks A LOT like the tumor does? THE FREAKING TUMOR!!!!!! Absolutely poetic that $MRNA produced the EXACT HR benefit as neoadjuvant pembro did. Truly, the most hilarious possible outcome. Now can we move on from this bullshit? $MRK and $MRNA should feel free to study their vaccine against the REAL standard of care now - NEOadjuvant immunotherapy. From my perspective, this has been total fugaze, and an amateurish pump and dump worthy of much lesser enterprises.

RAS(ON)i daraxonrasib in pretreated RAS mutant (non G12C) lung cancer, high grade tox in 54% incl 2.9% deaths. Tumor responses in 31-37% of pts, PFS 8.3m. So far not the breakthrough in lung cancer that it has been in pancreatic cancer #lcsm
In a phase 1–2 study of daraxonrasib, 54% of patients with RAS-mutant non–small-cell lung cancer had grade 3 or higher adverse events, mainly pneumonia, diarrhea, and rash; more than 30% of patients had a response. Full study results: nej.md/4h0GHUW

🧬 Could the future of cancer immunotherapy be multi-target instead of single-target? In this fascinating @ProgressPotent1 conversation, I sit down with Dr. Jack Silberstein - @Jack_Silb - Founder and CEO of Deck Bio, to explore a next-generation approach to T cell engager therapies designed specifically for solid tumors. We discuss: 🔹 Why traditional T cell engagers succeeded in blood cancers but struggled in solid tumors; 🔹 The biology of peptide-MHC (pMHC) targeting; 🔹 Tumor heterogeneity and antigen escape; 🔹 Multi-target immune recognition; 🔹 Deck Bio’s AACR 2026 preclinical data; 🔹 The future of engineered TCR therapies and combinatorial cancer targeting; Deck Bio’s core thesis is fascinating: 👉 Cancer may not be a “single antigen” disease - and next-generation immunotherapies may need to recognize patterns of tumor identity instead of one marker at a time. A deep dive into one of the most technically ambitious areas in modern oncology and immune engineering. #CancerResearch #Immunotherapy #DeckBio #TCellEngagers #Biotech #AACR2026 #Oncology #TCRTherapy #PrecisionMedicine #CancerInnovation @russopartners

Using our solved structure of LAG-3 dimer, we mutated the dimer interface to reveal dimerization as key to LAG-3 function. More importantly, we uncovered the mechanism of the LAG-3 antibody C9B7W, which binds a distinct site from clinical LAG-3 antibodies... 2/3

...and is the main antibody used by biopharma to validate LAG-3 as a therapeutic target. We found this antibody binds to domain 2 and blocks LAG-3 function by disrupting the LAG-3 dimer. We hope our findings will contribute to the development of improved LAG-3 therapeutics. 3/3
