Three in five human proteins are intrinsically disordered — no fixed shape, long written off as "undruggable." Congrats to the @ToposBio team on Topos-2 (new state of the art on PeptoneBench) and Topos-Bind, the first model to capture how these proteins shift shape when bound to a small molecule. Proud investor.
Today we are announcing Topos-2, our next-generation model for generating the full range of shapes a protein can take, across the order-disorder spectrum. We are also previewing Topos-Bind: to our knowledge, the first generative model to capture the range of shapes for disordered proteins bound to small molecules. Today, @autopoiesislab announced their $11M Seed round led by @thresholdvc. Autopoiesis is building AI capable of autonomous scientific discovery - moving AI from assisting scientists to discovering new knowledge itself. We believe AI can fundamentally change the pace of scientific progress, and feel privileged to partner with Joseph, Eike, Larry and the brilliant team building toward that future. Most model trainings have failed outside of frontier labs. Even inside frontier labs, knowing how to train for very different capabilities is often a matter of taste. Today, we introduce AutoScientist by @adaption_ai which sets out to change that.
Introducing AutoScientist. Most model training fails outside of frontier labs. AutoScientist automates the full research loop so it doesn't have to.

The pain, mess, and fulfillment of reinventing a company from software to AI. I hope it helps other leaders in the trenches doing the same.
Late last year, I found myself wondering whether any pre-AI companies had actually made the leap to AI-native and started asking around. One name came up over and over again: Notion So I teamed up with my longtime bestie and former Notino @camillericketts to take a look inside. The product has new energy and the business is booming, but the thing that caught our attention was the culture that turned the key on it all. Here’s what we saw inside a decade old company reborn to meet the AI revolution. Our latest company dispatch on @NotionHQ for @colossusmag

Proud to share that @thresholdvc has invested in @adaptionlabs, which is announcing a $50M seed today. I couldn't be more excited to partner with @sarahookr and @sudip_r0y as they build (at lightning speed!) fundamentally different AI systems - models that are highly efficient and adapt in real time. Congrats to the entire Adaption Labs team on this milestone. Even more exciting things coming very soon - stay tuned! 🚀

If this sounds a bit different than my usual podcast it’s literally because I thought Nick and I were just chatting pre-recording. Nope, that chat was the podcast. Probably made it better.
I spoke w/ @cloudflare co-founder & CEO Matthew Prince (@eastdakota) for the latest ep of the Slow Hunch podcast. Since 2010, Matthew and his team have built Cloudflare into one of the most important companies on the internet: powering and protecting vast portions of global traffic. I’ve known Matthew since @USV’s investment in Cloudflare’s Series C back in 2013, and it was really fun tracing his slow hunch, right from those early days. We covered: - how he met co-founders Michelle Zatlyn (@zatlyn) and Lee Holloway (@icqheretic) - the early experiments and risks that shaped Cloudflare - stories from the company’s pre-IPO days - their decision to make website encryption free - how their infra ended up running 2 of the internet’s 13 root servers - his thoughts on AI (esp the transition from a search-driven internet to an answer-driven one) Hope you enjoy!

When Charlie Kirk invited me onto his show, I had a lot of hesitation. I knew little about him and had heard mixed opinions. I watched a few episodes and saw a decent guy who I often disagreed with but who was respectfully searching for truth. Charlie had read my book and wanted to talk about it. I agreed to come on under the premise that we wouldn't discuss politics. We had a terrific conversation. Charlie was fascinated by taking cold showers, stoicism, mindfulness—the deeper questions of how to live well. He seemed like a genuinely good guy who cared about more than just politics. What struck me most was this: Charlie understood that free speech isn't singing kumbaya with people who agree with you. It's about having conversations with those you vehemently disagree with. He made a career out of it—willing to hear and debate anyone. When I was growing up in the Soviet Union truth was only whispered in the privacy of small kitchens. You could lose your job, be sent to jail, or worse, to the gulag for reading the wrong books or holding the "wrong" opinion. I understand what real oppression looks like—I lived through it. I once thought America was different—a place where people could disagree, debate, and criticize openly. But that America is fading. Charlie understood this. He believed in the marketplace of ideas, even when—especially when—those ideas challenged his own. Today America became less free. Rest in peace Charlie.

There's so much money being sloshed around right now (esp for AI startups). One simple thing founders can do is actually take the money (with minimum dilution), but then don't feel the urgency to spend it all. Don't chase the next round, chase customers instead.
Timely repost, "Raise less, build more" trohan.com/2023/08/20/raise-… Why VC funds offer founders huge checks. Why VC raise huge funds. Why giving startups huge checks isn’t empirically better. How certain companies like Notion, Vanta, Linear and Zapier are paving a new way.

I'd share a coffee and chat with @mollywood any day. This time just involved a camera and was recorded :-). Continue to be grateful to work with the @thresholdvc team and our amazing founders. We believe in "long-term right" and are actively investing in early-stage companies.
.@emelton explains the tricky situation that first-time fund managers who raised & invested at the market peak (2020-2021) find themselves in: "Some (new VCs) WILL be poor performers. The question is, over what period of time do you judge them?"
