Quick takes on the Social Signals from #ASCO26. 🧬 RASolute-302: @marklewismd 's tweet regarding the standing ovation for RASolute-302 really made that trial stand out. The momentum here was massive, fueled by high expectations from the @RevMedicines PR, #AACR26 presentations, and the high-profile Ben Sasse interviews. ⚔️ PROTEUS: One of the more interesting trials from a social media perspective, as the radiation oncology community on X was highly critical of the trial's surgical paradigm. 🔄 CROWN: Felt a bit like an incredible replay of the Plenary from years ago—steady, practice-confirming data. 🎬 HARMONi-6: Perhaps the most compelling narrative twist. The discussant, Dr. Julie Brahmer, masterfully set up the Plenary audience using The Devil Wears Prada as a research metaphor. It begs the question: Is VEGF back? 🚀 LIBRETTO-432: A massive surprise for many attendees. Lung cancer specialists were thrilled about @LillyOncMed selpercatinib in the adjuvant setting for RET-positive NSCLC. The core takeaway from @christine_lovly presentation was loud and clear: "This is why we test." 🔬 OPTIMA: Really got the breast cancer community excited about using @Veracyte 's test to safely reduce the chemotherapy burden for many of their patients. 📦 WU-KONG28: Had such an interesting vibe. Great data from @Dizal_Global , but it leaves the KOLs asking the same commercial question: Why isn't this readily available to patients in the US yet? My thoughts from the #ESMO26 conference on thoracic oncology 1️⃣ T-cell engagers are changing SCLC — DeLLphi-305's 58% mortality reduction in 1L maintenance and DeLLphi-304's mature, approved relapsed benefit. 2️⃣ ADCs and rational EGFR/MET combinations are expanding post-resistance options — but watch the fine print: China-only populations, interim OS, and (for PANKU) heavy myelosuppression. 3️⃣ Perioperative IO and ctDNA are reshaping early-stage care — NEOTORCH is OS-positive at final analysis; NeoCOAST-2 and MDT-BRIDGE show ctDNA and MDT-adaptive strategies worth testing in phase III. An outstanding year for thoracic oncology — with the usual reminder: interim OS, single-arm designs, and regional populations temper the headlines.

Final analysis of DREAMM-3 #mmsm @BloodCancerJnl ✅ As monotherapy, belantamab is not better than Pom/Dex (no difference in PFS, PFS2 and OS) even with longer follow up ✅ About 20% (24 of 122) of patients who had a first grade ≥2 eye exam finding still hadn’t recovered from it at their last visit ➡️ rdcu.be/rypZoXm45vba

Doctors read these CT scans as normal. An AI model looked at the same scans and flagged pancreatic cancer anyway. In a study published in Gut, researchers took scans from people who were later diagnosed with pancreatic cancer and compared them with scans from people who stayed cancer free. The AI picked out the cancer on about 73% of the earlier scans. Experienced radiologists caught about 39%. On average the AI saw something about 16 months before the diagnosis. The catch: this looked backward at old scans. It still needs testing going forward, in people at higher risk, before anyone uses it on patients. Study: pubmed.ncbi.nlm.nih.gov/4204…

This is also true of pharma trials.
Our tax dollars fund unethical trials with substandard control arms for patients with cancer. Please read this paper in Nature Reviews (this is not behind a paywall) drive.google.com/file/d/1txu…

Great paper on benefit of Tam-peli easily beating SOC topotecan in 2nd line ES-SCLC…. But wrong figures attached by NEJM on sac-TMT..now fixed it! (Guess it is not only me struggling w ADC nicknames…) Excellent data by Dr Zhao et al and can hardly wait to see one of these agents approved as study also shows sobering new reality for us- w tarla moving into maintenance and lurbi “ousted”, are we supposed to go back to topotecan’t (as famously coined by Dr. Socinski? @CoreyLange28917 )? With pitiful data as seen in this study: 60% of pts progress on topo by 3 months, ORR<10% and with grade 3 or higher AEs at 75% (!) and very burdensome schedule i just can’t…and our patients shouldn’t.. #lcsm

The #SABCS26 abstract titles are out! One study that particularly caught my attention is STOP-HER2, which will be presented in GS3. These results could provide crucial insights into de-escalation strategies for exceptional responders to anti-HER2 therapy. Looking forward to the data! sabcs.org/events/
Had a wonderful and inspiring time at #ESMO26! Honored to give a Rapid Oral Presentation, have our paper published simultaneously @ESMO_Open and receive an ESMO Merit Award. Truly grateful to all my mentors and collaborators! @myESMO @PTarantinoMD @NateraGenetics Also had a great meeting with the SOLTI Young group on the final evening. Looking forward to future collaborations! @_SOLTI @Rodrosb @shigehira @YukinoriOzaki @9p2nNkRpjOrdvcw @ToshimiHBM @NateraOncology

People who have been in research fields develop a felt sense that most "research" (in almost any field that isn't exactingly technical) is research theater, and many superstar careers enjoy star status not because of a unique connection to the truth but because of a unique connection with an audience. No one really has the time or stature to, from a position of objectivity and trust, bring popular researchers to heel. But it will get increasingly hard to hide from serious critique. The machines can do critique now, at the level of depth and rigor previously only possible for the academically deranged. We're going to have to revise a lot of what we "know" about the world that isn't really knowledge but is actually just received popular consensus - like floating voltages without grounding wires. The grounding wires are coming.
Tyler Cowen: 'I am not even sure what to say at this point. We need a radical revaluation of this work, and of the associated policy arguments. More generally, this is only a single step in the story of how AI is going to revise economic science, and indeed past research more generally. As I stress in many of my recent talks, there will be significant gains and losses in status, with the losses often outweighing the gains. I see this as one of those cases.'
