Agree, lung cancer ADC/BsAb pipelines look the same. We mapped 47 active trials (@ClinLungCancer): • BsAbs ~70% / ADCs ~30% • Top BsAb: EGFR×c-MET (~28%) • Top ADC: TROP2 (~33%), HER2 (~20%) Same few antigens. More me-too than novel. doi.org/10.1016/j.cllc.2024.…
Since people are talking about target crowding, here's a chart from an upcoming post: <5% of new drugs entering the clinic are going after targets that haven't been tested clinically before, ~60% are going after targets with an approved drug. New ideas are hard to find!

🔥 Outstanding piece of work published today in Nature Medicine! Huge congratulations to @MartinReck2 ,@NadalErnest , @DoctorJSpicer and @AndrearicFili , and the entire MDT-BRIDGE team on this important publication! 👏👏 The phase II MDT-BRIDGE trial, investigating neoadjuvant durvalumab plus chemotherapy in resectable or borderline resectable NSCLC, tackles a clinically relevant question: how can we push the boundaries of multidisciplinary treatment and offer more patients a chance of curative-intent therapy? This is exactly the kind of collaborative clinical research we need: challenging established boundaries, asking meaningful questions, and ultimately striving to improve outcomes for our patients. Congratulations to everyone involved! 👏 🔗 nature.com/articles/s41591-0… Happy to continue sharing exciting advances in lung cancer research as an #ESMOAmbassadors #ESMO26 #NatureMedicine #LungCancer #NSCLC #ThoracicOncology #Immunotherapy #ClinicalResearch #MultidisciplinaryCare #CancerResearch

Huge milestone for improving the treatment of extensive stage SCLC. Feeling happy and hopeful for our patients. Huge congratulations to the entire clinical trial team, to Amgen, to the study investigators and the excellent presentation and discussion by @sands_jacob @StevenLin_MDPhD The work continues - let’s make sure tarlatamab gets wide access, and other DLL3 T cell engagers get incorporated into the treatment paradigm (in clinical trials), and let’s find better solutions for those DLL3 T cell engagers don’t work in.
Outstanding discussion by @StephenVLiu on the practice-changing DeLLphi-305 trial @sands_jacob . A new chapter in the treatment of ES-SCLC! #ESMO26. One year ago, we anticipated that we might finally be bending gravity. Today, that vision is becoming a reality!!!!!!👏👏👏👏👏

Game changing data. Congratulations to the team and gratitude to the patient participants and their families. #SCLC @LUNGevity
#ESMO26 Presidential 1 : #SCLC DeLLphi-305: Who would have imagined some years ago that median OS of SCLC would equal that of non-AGA NSCLC! #Tarlatamab as 1L maintenance with Durvalumab after induction chemoIO in ES-SCLC improved OS from 12.7 to an impressive 21.1m, corresponding to a HR of 0.42. 80% of patients alive and 38% progression-free at 12 months The dawn of a new era for thoracic oncology and for the lives of our patients 💫✨ @myESMO @SclcSMASHERS #ESMOAmbassadors

The modern day “ fortune teller” ?:) jokes aside I think these AI models do have lots of potential.
There's a lot in a face photograph about health that AI can detect, as seen in this foundation model of >40 million face images, including outcomes from cancer, biological age, influence of lifestyle factors, and survival @LancetDigitalH thelancet.com/journals/landi…

We now have prospective data with TOPO1 ADC sequencing from: - SATEEN (#ESMOBreast26) - HER3-DXd (#ESMOBreast26) - SWITCH (upcoming #ESMO26) All validate prior retrospective data, showing that TOPO1 ADC resistance is not overcome by target switch. New payloads urgently needed.
#ESMO26 | SWITCH – Phase II In mBC progressing after ADC therapy, switching the target antigen while continuing with the same TOP1 inhibitor payload class resulted in ORRs of only 0–8.3%. Although the sample size was small and the patient population heterogeneous, changing the target antigen alone does not appear sufficient to overcome resistance.

🚨 Hot off the press! A week before @myESMO annual meeting for everyone to read 👉 🔥Pleased to share with the 🌎our comprehensive review in @CellPressNews @MedCellPress journal Med: 👉“Cancer plasticity and histological transformation as adaptive resistance mechanisms in precision oncology” 👉Precision oncology has transformed cancer therapy, but tumors adapt. 👉One of the most striking escape routes is histological transformation: a cancer that responds beautifully to targeted therapy can change its very identity, from adenocarcinoma to small cell or squamous for example. 👉In this review we examine: 🔹 How different forms of tumor plasticity drive histological transformation as a resistance mechanism across anticancer therapies 🔹 The underlying biology + mechanisms behind these lineage switches 🔹 How precision oncology, informed by a deeper understanding of plasticity, can be used to anticipate, detect + overcome transformation + improve outcomes for patients 👉If we want the next generation of cancer therapies to last, we have to treat cancer as an evolving system, not a static target.
