Great Results from DeLLphi-305 🫁 however with CRS in nearly half of patients, this means readiness is the real challenge now. Monitoring pathways, step-up dosing logistics and training community teams will decide how quickly this reaches patients outside academic centres. #LungCancer
DeLLphi-305🚨 A 0.42 hazard ratio for death in extensive-stage small-cell lung cancer. In a disease where we’ve celebrated gains of two to three months, this is a different level. 🫁 Tarlatamab–Durvalumab Maintenance for Extensive-Stage Small-Cell Lung Cancer nejm.org/doi/full/10.1056/NE… @MP_3855, @ADesaiMD 📊 In the open-label phase 3 DeLLphi-305 trial, 563 patients with extensive-stage SCLC without progression after first-line chemoimmunotherapy were randomized to tarlatamab plus durvalumab or durvalumab alone as maintenance. The median overall survival was 21.1 versus 12.7 months (HR 0.42), and the median PFS 7.4 versus 1.9 months (HR 0.34). Grade ≥3 adverse events occurred in 46.8% versus 40.6%; cytokine release syndrome, mostly grade 1–2, affected nearly half of tarlatamab-treated patients. #LungCancer Pinging our faculty @christine_lovly, @GlopesMd, @DrSanjayPopat, @StephenVLiu, @jillfeldman4, @UOzkerim, @DrMirallas, @MartinReck2 , @bmassutis @UmbertoMalapel1 , @CharuAggarwalMD @peters_solange, @HenningWillers, @HHorinouchi, @BRicciutiMD, @PercyLeeMD, @bensolomon1, @Latinamd, @DrJNaidoo, @barlesi, @VanitaNoronha, @ChristianRolfo @Tony_Calles @s_couraud @marinagarassino @g_mountzios @BairdAM @barlesi @JFreemanDaily @Piuchagarrido @PercyLeeMD @bensolomon1 @VanitaNoronha @alexdrilon @JordiRemon @JulienMazieres @TerriConneran @HenningWillers

🔬 Can adding an androgen receptor inhibitor to ADT and radiotherapy improve metastasis-free survival in very high-risk localised prostate cancer? New Phase 3 interim results from DASL-HiCaP suggest a significant benefit. 📊 LBA4 – DASL-HiCaP (ANZUP1801): Darolutamide with ADT and definitive or salvage locoregional radiotherapy cslide.ctimeetingtech.com/es… Darolutamide added to ADT and locoregional radiotherapy significantly improved metastasis-free survival in patients with very high-risk localised or biochemically persistent/recurrent prostate cancer. At four years, metastasis-free survival was 95% versus 88% with placebo (HR 0.42; p=0.0001). ✅ These findings support treatment intensification, although longer follow-up is needed to establish whether this benefit translates into improved overall survival. #ProstateCancer @niazi_Dr7, @Prof_IanD, @ChrisSweens1, @AlisonZhangMBBS, @seanmmcbride, @DocJarad, @DavePryorRO, @MartinStockler @OncoAlert 🚨 #LPCCC27 @Silke_Gillessen @AOmlin @ProfKHerrmann @weoncologists

🔬 Can perioperative immunotherapy plus an antibody–drug conjugate change the treatment landscape for cisplatin-ineligible muscle-invasive bladder cancer? New Phase 3 data suggest it could. 🧬 Durvalumab, Tremelimumab, and Enfortumab Vedotin in Muscle-Invasive Bladder Cancer 🔗 nejm.org/doi/full/10.1056/NE… 📊 The Phase 3 trial demonstrates significant improvements in event-free survival with perioperative durvalumab plus neoadjuvant enfortumab vedotin, with or without tremelimumab, compared with radical cystectomy alone in cisplatin-ineligible or cisplatin-declining patients with muscle-invasive bladder cancer. The durvalumab–enfortumab vedotin combination also significantly improved overall survival. ✅These findings highlight a promising perioperative treatment strategy, although the substantial incidence of grade 3 or higher adverse events warrants careful consideration. @tompowles1, @drenriquegrande, @Uro_Oncologist, @TimuinL, @HernanJCutuli, @DrRosenbergMSK @OncoALert 🚨
